The difference in ipamorelin vs sermorelin is that sermorelin is a GHRH receptor agonist and ipamorelin is a ghrelin receptor agonist. Sermorelin copies residues 1 to 29 of human growth hormone releasing hormone, amidated and otherwise unmodified, and signals through cyclic AMP. Ipamorelin is a synthetic pentapeptide that binds GHS-R1a and signals through calcium. Sermorelin models the hypothalamic signal in its simplest active form, ipamorelin imitates a gastric hormone, and a study selects between them by the receptor it intends to probe.

Is Sermorelin a GHRH or a GHRP?
Sermorelin is a GHRH analogue, the native 1-29 amide fragment of growth hormone releasing hormone, not a GHRP. Every compound in this area of peptide research falls on one side of a simple divide. Releasing hormone analogues act on the GHRH receptor. Growth hormone releasing peptides and their descendants act on the ghrelin receptor.

Pituitary models respond to both classes, and to the combination more strongly than to either, because the two receptors converge on the same somatotroph through separate intracellular routes. The ghrelin receptor also operates in the hypothalamus, where its activation appears to lower somatostatin restraint and so makes the pituitary more responsive to available GHRH.
What Is Sermorelin?
Sermorelin is the synthetic 1-29 amide fragment of human GHRH, the shortest sequence that retains the full biological activity of the 44 residue hormone. The natural hormone was first purified in 1982 from a pancreatic tumour that secreted it in excess, and the 1-29 fragment was soon shown to carry its activity. Sermorelin in 5mg and 10mg vials is made by solid-phase synthesis: Tyr-Ala-Asp-Ala-Ile-Phe-Thr-Asn-Ser-Tyr-Arg-Lys-Val-Leu-Gly-Gln-Leu-Ser-Ala-Arg-Lys-Leu-Leu-Gln-Asp-Ile-Met-Ser-Arg-NH2, molecular weight about 3,358 daltons.
Its defining property is instability. The alanine at position 2 makes it a substrate for dipeptidyl peptidase-4, which removes the first two residues and leaves an inactive chain, so the fragment persists only minutes in serum or plasma. For comparative work that is an advantage: sermorelin shows how the unmodified hormone sequence behaves, so any added persistence seen with an engineered analogue can be attributed to the engineering. It was formerly marketed in the United States as a diagnostic product, and that commercial supply ended in 2008.
What Is Ipamorelin?
Ipamorelin is Aib-His-D-2-Nal-D-Phe-Lys-NH2, a 712 dalton synthetic pentapeptide designed as a selective ghrelin receptor agonist. Ipamorelin 10mg descends from the GHRP series of the 1980s and 1990s but was optimised for selectivity. In the 1998 swine study that introduced it, ACTH and cortisol held steady as growth hormone climbed, unlike the response to older releasing peptides, and prolactin was unaffected. Three unnatural residues make it far more resistant to peptidases than a natural pentapeptide.
How Large Is the Molar Gap Between the Two?
The molar gap is close to fivefold, because ipamorelin is so much smaller than sermorelin. A milligram of sermorelin contains about 0.30 micromoles of peptide, while a milligram of ipamorelin contains about 1.40 micromoles. Weight-matched comparisons are therefore not molecule-matched, and receptor response studies should set concentrations in molar units.

Where Does CJC-1295 No DAC Fit Between Them?
CJC-1295 no DAC sits on the sermorelin side of the divide, as the same 29 residue GHRH fragment with four stabilising substitutions. CJC-1295 without DAC, also sold as modified GRF 1-29, carries D-Ala2, Gln8, Ala15 and Leu27. The first blocks DPP-4, and the others reduce cleavage and oxidation-prone sites elsewhere in the chain. Its mass rises only slightly, to about 3,368 daltons, which makes sermorelin the ideal control for it: same length, same receptor, four residues apart.
The same logic underlies the CJC-1295, ipamorelin and sermorelin stack that appears in search data. As a research design, the three compounds form a coherent set. Sermorelin supplies the native GHRH baseline, CJC-1295 no DAC the stabilised form of that signal, and ipamorelin the second receptor. Laboratories that want the stabilised pair pre-mixed can use the CJC-1295 and Ipamorelin 10mg blend and keep sermorelin in a separate vial as the unmodified comparator.

All three peptides dissolve readily in bacteriostatic water containing 9 mg per mL benzyl alcohol. Divide milligrams by millilitres for the strength; each U-100 graduation equals 0.01 mL.

Which Compound Should a Study Choose?
A study should choose sermorelin when it needs the native GHRH sequence and ipamorelin when the question concerns the ghrelin receptor. Our recommendation is to order sermorelin alongside any engineered GHRH analogue as the native control, because without it there is no way to show what the modifications changed. Add ipamorelin to a GHRH analogue only after each compound has been characterised alone.
