BPC-157 vs TB-500: What Is the Difference?

The difference between BPC-157 and TB-500 is that they are unrelated molecules: BPC-157 is a synthetic 15 amino acid peptide of about 1,419 Da derived from a gastric juice protein, while TB-500 is synthetic material based on thymosin beta 4, a 43 residue actin-sequestering protein of about 4,963 Da. BPC-157 is studied mainly for VEGFR2, focal adhesion kinase and nitric oxide signalling; thymosin beta 4 for actin dynamics and cell migration. They share no sequence and act through separate pathways.

What Is BPC-157?

BPC-157 is a synthetic pentadecapeptide, Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val, taken from a larger body protection compound isolated from gastric secretions in the early 1990s. A run of three prolines near the N-terminus gives it an unusually rigid backbone for so short a chain, and reports describe it remaining intact at a low pH that would fragment most chains of similar length.

Mechanistic work concentrates on signalling. A 2011 Journal of Applied Physiology study in tendon fibroblasts linked the peptide to the focal adhesion kinase and paxillin pathway, which governs cell adhesion and spreading. A 2017 Journal of Molecular Medicine study in endothelial cells reported VEGFR2 activation, an early step in new vessel formation, and many rodent studies examine its interaction with the nitric oxide system. A pharmacokinetic study in rats and dogs, published in Frontiers in Pharmacology in 2022, reported a short plasma half-life for the intact peptide. What Is TB-500?

TB-500 is the research-market name for synthetic material based on thymosin beta 4, a 43 residue protein with an acetylated N-terminus that is present in nearly every mammalian cell and abundant in platelets. Within the cell it regulates the actin pool: monomeric G-actin is bound one to one and held back from filament assembly until required, which makes the protein a determinant of cell shape and motility. A 1991 Journal of Biological Chemistry report identified it with Fx, an actin-sequestering peptide first isolated from platelets, and binding depends on the central motif LKKTETQ.

Some suppliers apply the name to the full chain and others to a short fragment built around the actin-binding motif, so the certificate for each lot of our TB-500 vials states which form is present. The best-known studies, a 1999 rat dermal model in the Journal of Investigative Dermatology and a 2004 Nature paper linking the protein to integrin-linked kinase in cardiac cells, used the full-length molecule.

How Do BPC-157 and TB-500 Differ in Structure and Mechanism?

BPC-157 and TB-500 differ in size, in where they act and in the research questions each is used to address, which is why they are usually treated as separate experimental variables.

Size and Structure

At roughly 1.4 kDa, BPC-157 has under a third of the mass of full thymosin beta 4. A shorter chain is simpler to synthesise and to verify by mass spectrometry. A longer one presents more sites for oxidation and cleavage, and the methionine at position 6 of thymosin beta 4 is a recognised oxidation point, one reason that material is best kept cold and dry until reconstitution.

Site of Action

BPC-157 studies centre on signalling at and around the cell surface, with VEGFR2 and focal adhesion kinase the recurring targets. Thymosin beta 4 acts largely inside the cell on the actin pool, although extracellular roles have also been reported in migration assays.

Research Applications

Laboratories select BPC-157 for vessel formation, tendon cell and gut lining models, and thymosin beta 4 when the question concerns cytoskeletal dynamics, cell migration or the actin cycle. Can BPC-157 and TB-500 Be Combined in One Vial?

Yes, BPC-157 and TB-500 can be combined in one vial, and they are manufactured as a co-lyophilised pair because both are water-soluble and reconstitute in preserved diluent for peptide stocks. The substantive question is experimental design, and there are three ways to set it up.

For separate vials, the recovery stack of two 10mg vials supplies one of each under its own lot number. For a single reconstitution, the BPC-157 and TB-500 blend in 5+5mg or 10+10mg holds equal amounts of both. Adding 2 mL to the 10+10mg vial gives 10 mg per mL combined, half of it each compound, so one U-100 unit carries 50 mcg of each.

Combining stocks by hand is also possible, but the arithmetic changes. Each solution dilutes the other, so equal 1 mL portions of two stocks each prepared at 5 mg per mL combine into 2 mL holding 2.5 mg per mL of each. Both lot numbers belong on the new vial.

Should a Study Use a Single Compound or the Pair?

A study should use single-compound vials whenever an observed effect must be attributed to one molecule, and add the pair only when the combination itself is the subject. A design comparing each peptide alone with both together has three arms, so it requires single vials as well as the combination.
